Sulphide is one of the known environmental stressors, which potentially binds to cytochrome C oxidase (COX), a key enzyme in the electron transport chain, thereby blocking oxygen transport and ATP production. To ascertain the toxic effects of sulphide on Pacific white shrimp, Penaeus vannamei, two distinct exposure experiments were carried out with varying concentration of sulphide (0–1 mg/L), dissolved oxygen (normoxia and hypoxia) and pH values (8.2, 6 and 5). The activity of enzymes viz COX, superoxide dismutase (SOD), phenoloxidase and lactate accumulation was investigated. Outer membrane integrity and COX monomer separation were also done with the isolated crude mitochondrial preparations. Results indicated a significant reduction (p ≤ .05) in COX enzyme activity in sulphide exposed treatments when compared to control. The reduction was more intense when pH levels were reduced under hypoxia condition. Lactate accumulation as a result of anaerobic metabolism was found to be higher in hypoxic treatments. No significant difference (p ≥ .05) was observed in superoxide dismutase activity between the treatments, whereas phenoloxidase activity significantly decreased at higher concentration sulphide. Separation of mitochondrial proteins resulted in the identification of ~205 kDa of COX monomer, and significant damage was found in outer membrane integrity under hypoxia and pH treatments. From this study, it is evident that at a given concentration, sulphide is toxic to P. vannamei, and in association with hypoxia and low pH, they further intensify sulphide toxicity. Our results indicated that sulphide toxicity should not be considered as a single factor, rather it should be a considered as combination of factors.  相似文献

Artemia culture in the Mekong Delta, Vietnam, is becoming more and more important to satisfy the global demands of cyst production. Nevertheless, Artemia cyst productivity in the Mekong Delta has been fluctuating largely due to not only technical inefficiencies and external factors but also farms’ characteristics. This study employed a data envelopment analysis to evaluate the production efficiencies in correlation with biological and cost inputs of Artemia culture in Vinh Chau and Bac Lieu located in the Mekong Delta. The key factors affecting technical efficiency (TE) were determined using ordinary least squares regression model. The empirical results indicated that production efficiencies of farms in Vinh Chau and Bac Lieu were similar with metatechnology ratios of 0.97 and 0.96 respectively. About 62.8% of surveyed farms were operating in technically inefficient circumstances. Farmers’ experience, educational level and culture technique training participation had positive effects on TE, while climate effects and water problems have been proved to have negative effects on TE. To improve production efficiency, inefficient farms are suggested to have moderately increasing combinations of organic fertilizer, chemical and feed. The use of probiotics should be reduced as it increases production cost and affects TE. Notably, a collaboration of scientists and feed processing companies is recommended to produce a specific formulated feed for Artemia in commercial scales. This study also proposes adaptive policies for local governments and cooperatives to assist coastal famers in the Mekong Delta overcome technical inefficiencies, improve cyst productivity and obtain economic sustainability in Artemia culture.  相似文献

为探究马氏珠母贝肉酶解产物(Enzymatic hydrolysate from Pinctada martensii,EP)对酒精性肝损伤(Alcoholic liver damage,ALD)的保护作用,该研究将EP超滤分级为截留分子量>10 kD(EP-Ⅰ)、3~10 kD(EP-Ⅱ)和<3 kD(EP-Ⅲ)3个组分,检测其体外抗肝损伤活性及对ALD小鼠肝保护作用的影响。体外试验结果显示,EP-Ⅲ可显著激活体外乙醇脱氢酶(ADH)活性(P<0.01),3个超滤组分均具有一定的体外抗氧化能力且EP-Ⅲ>EP-Ⅱ>EP-Ⅰ;动物试验结果显示,与模型对照组相比,各超滤组分均能够显著降低小鼠血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)活力、小鼠肝脏指数及肝脏中丙二醛(MDA)和甘油三酯(TG)含量,同时显著增强小鼠肝脏中超氧化物歧化酶(SOD)、乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)活力,提高肝脏中的谷胱甘肽(GSH)含量。综上,马氏珠母贝肉酶解超滤组分对急性ALD具有一定的辅助保护作用,其中EP-Ⅲ的保护作用效果最佳,其机制可能与加快机体乙醇代谢和减缓乙醇对机体造成的氧化损伤相关。  相似文献

An 8‐week feeding trial was conducted to evaluate the effects of dietary copper sources on growth performance and intestinal microbial communities of juvenile Pacific white shrimp (Litopenaeus vannamei). Four isonitrogenous (420 g/kg crude protein) and isolipidic (70 g/kg crude lipid) experimental diets were formulated to contain different copper sources, Diet NSC (no‐supplemented copper), Diet CS (copper sulphate), Diet Availa (copper amino acid complex from Availa®Cu100) and Diet M (1:1, copper sulphate + copper amino acid complex). A total of 360 Pacific white shrimp juveniles (initial weight: 1.86 ± 0.03 g) were randomly allocated into four groups, with three replicates per group and 30 shrimps each replicate. An Illumina‐based sequencing method was used to analyse the intestinal bacterial composition of shrimp juveniles. The results revealed that shrimps fed Diet M had significantly higher percent weight gain (PWG) than that fed Diet NSC and Diet CS, while there were no significant differences between Diet M and Diet Availa. Survival was not affected by the dietary copper sources. Proteobacteria and Bacteroidetes were dominant in the intestines of Pacific white shrimp. Shrimps fed Diet M and Diet Availa had higher bacterial richness and diversity than those fed Diet NSC and Diet CS with no significant differences among all treatments. Furthermore, Diet M and Diet Availa which contained copper amino acid complex reduced the relative abundance of Vibrionaceae in shrimp intestinal tract. This study indicated that dietary copper sources could affect the intestinal microbial communities and diets supplemented with Availa®Cu100 could have a positive impact on the growth performance and decrease the potential risk of disease in Pacific white shrimp.  相似文献

The present trials tested the efficiency of surplus spermine to reduce inflammation and oxidative stress following LPS‐induced stress using an in vitro model of head kidney and liver cells isolated from Atlantic salmon. Spermine did not protect cells from LPS‐induced inflammatory response at either 0.3, 0.6 or 0.9 mM. However, as the gene expression of spermidine/spermine N1‐acetyltransferase (SSAT) increased with increasing spermine concentration, we addressed possible oxidative effects of the increased SSAT using its activator DENSPM or inhibitor of polyamine oxidation of the acetylated polyamines using MDL72527 at a spermine concentration of 0.6 mM. There was no significant effect of DENSPM, but MDL72527 decreased gene expression of GPX‐3 (p = .04), while gene expression of catalase and MnSOD was unaffected by treatment (p = .30 and p = .48, respectively). In conclusion, spermine did not protect cells from LPS‐provoked inflammation. The higher the spermine concentration, the more SSAT producing acetylated spermine occurred. Inhibiting the acetylated polyamine oxidases by MDL72527 improved oxidation status as expected due to a lower endogenous production of H2O2 by polyamine and acetylated polyamine oxidases. Probably care should be taken using polyamines or arginine as functional ingredients to avoid any increased oxidation within cells.  相似文献

This study was conducted to evaluate the application of four fish origin probiotics to relieve the side effects induced by SBM in shrimp. Shrimps were fed with high fish meal diet (C0, positive control containing 500 g/kg FM and 0 g/kg SBM), low fish meal diet (C, control containing 250 g/kg FM and 340 g/kg SBM) or the control diet (C) supplemented, respectively, with 1.0 × 108 CFU/g Bacillus pumilus SE5 (T1), Psychrobacter sp. SE6 (T2), Enterococcus faecium MM4 (T3) or Bacillus claussi DE5 (T4). After 6 weeks of feeding, the growth performance, hepatopancreatic digestive enzymes, intestinal histological structure and immune parameters were determined. The results showed that compared with high FM group (C0), significantly lower WGR and SGR as well as higher FCR were observed in the low FM group (C) (p < .05). Compared with the control, significantly lower FCR was observed in treatment T1 (p < .05), but not in treatments T2–T4. Protease, amylase and lipase activities in all the treatments (T1‐T4) were significantly higher than the control (p < .05). GOT and GPT activities in the control were significantly lower than the C0 (p < .05), while higher MDA level was recorded in the control (p < .05). Meanwhile, higher GPT activities were observed in treatments T1‐T4 compared with the control (p < .05). Compared with the C0, lower SOD, ACP and AKP activities were observed in the control. Significantly improved SOD and AKP activities were observed in all probiotic feeding groups compared with the control (p < .05). The control diet led to significant reduction of intestinal wall thickness and villus height compared with the C0 (p < .05), while significantly higher intestinal wall thickness and villus height were exhibited in all the probiotic groups compared with the control (p < .05), except intestinal wall thickness in treatment T3. Thus, these results demonstrated that the four fish origin probiotic strains could relieve the side effects induced by high level of SBM in shrimp, while probiotic B. pumilus SE5 showed the best performance.  相似文献

A study was conducted to characterize the effects of dietary oxidized fish oil on the growth performance, immunity and antioxidant status of genetically improved farmed tilapia (Oreochromis niloticus) and to determine the role of ferulic acid on the oxidative damage induced by the oxidized fish oil. The tilapia (13.73 ± 0.31 g) were fed four experimental diets containing untreated (peroxide value, POV: 2.2 meq/kg) and highly oxidized (POV: 120.6 meq/kg) fish oil either with or without ferulic acid (0 or 400 mg/kg) supplementation for 12 weeks. From the results, the oxidized fish oil treatments increased antioxidant enzyme activities and MDA values but decreased the weight gain and the immunological parameters in tilapia. Meanwhile, the serum biochemical indices were significantly affected by the oxidized fish oil. Besides, the addition of ferulic acid partially counteracted the free radical‐induced damage and improved the health status of tilapia. In conclusion, the oxidized fish oil may induce oxidative stress, destroy liver, dysregulate lipid metabolism as well as reduce non‐specific immunity, and eventually result in growth inhibition of tilapia. The ferulic acid supplementation partially offset the negative effects of the oxidized fish oil on tilapia.  相似文献

This experiment was conducted to assess the effects of antimicrobial peptides at different levels (Diet 1 (0%), Diet 2 (0.1%), Diet 3 (0.2%), Diet 4 (0.4%), Diet 5 (0.6%) and Diet 6 (1%)), on growth, serum biochemical indices and antioxidant effect, feed utilization and disease resistance in Litopeneaus vannamei. There were four replicates in each group in the experiment (mean weight = 0.21 ± 0.00 g) and also fed with their respective diets for 8 weeks. Growth performance compared with the control group (0%) significantly increased at first and then decreased among treatment groups (p < .05) whereas the survival rate ranged from 78% to 96%. No significant differences were observed in terms of moisture, crude protein and ash content, but there was a significant increase in crude lipid (p < .05). In serum, acid phosphatase, alkaline phosphatase, polyphenol oxidase, triglyceride, glucose and total cholesterol changed as compared with 0%. Total antioxidant capacity, glutathione peroxidase, catalase, superoxide dismutase and malondialdehyde were also different from 0%. Disease resistance was increased in shrimp among treatment groups with 0.4% recording the lowest mortality percentage of 37% after the challenge test. The results from the present study suggested that supplementation of AMP at 0.4% in shrimp diet can improve growth performance, antioxidants activities and innate immune response of Pacific whiteleg shrimp.  相似文献

Agave lechuguilla is a succulent plant species, mainly distributed in the northeast of Mexico and south of the United States of America. The main use for this plant is the fibre´s extraction (known as Tampico fibres), resulting in 15% of fibres and 85% of a by‐product waste named guishe. The lechuguilla collectors, normally incinerate the guishe, thus causing environmental contamination. Interestingly, recent studies showed that guishe contains molecules with nutritional properties, such as saponins, flavonoids and sugars. Therefore, in this work, we evaluated the effect of the crude extract of guishe as a feed additive in whiteleg shrimp diets. According to that, MS‐HPLC analysis of the extract showed the presence of saponins such as diosgenin, smilagenin, hecogenin, manogenin, tigogenin hexose, yucagenin, chlorogenin, diosgenin diglucoside and the flavonol, quercetin. After chemical analysis, the crude extract was included into an experimental diet in four levels; 0% (L0%), 0.1% (L0.1%), 0.3% (L0.3%) and 0.6% (L0.6%). Dietary incorporation of the extract was evaluated by zootechnical performance, haemolymph biochemistry, histomorphology and digestive enzyme activity of shrimps. After 5‐week feeding, the L0.3% diet showed significantly higher growth and better feed utilization among treatments. A significant increase in tubule epithelium height and tubule coverage area from hepatopancreas in shrimp under L0.3% diet compared with the control diet suggest an improvement of the health and nutritional status of the shrimp. Inclusion of L0.3% and L0.6% of the crude extract resulted in a reduction in amylase activity, without effect in glucose levels in the haemolymph. Thus, we suggest that lechuguilla guishe crude extract contains nutritional molecules that may be used as a feed additive to promote shrimp productivity.  相似文献

The current study evaluates the efficacy of a low‐cost salt mixture (LCSM) to replace expensive reconstituted sea salt (RSS) in the salinity acclimation and nursery phase of Pacific white shrimp under laboratory and farm conditions. LCSM was formulated to yield sodium, potassium, calcium and magnesium concentrations closely comparable to that of diluted seawater. Laboratory‐based nursery trials were conducted at 2, 6 and 15 g/L salinities, incrementally replacing RSS with LCSM (25%, 50%, 75% and 100%) at four replicates per treatment. Thirty postlarvae were reared for 7 days in 24‐L aquaria during the 2 and 6 g/L trials, while the nursery trial for 15 g/L salinity was conducted for 21 days with 400 postlarvae stocked in 150‐L tanks. On‐farm evaluation of LCSM was carried out in two tank‐based systems installed on levees adjacent to shrimp production ponds. RSS was incrementally replaced with LCSM (0%, 50%, 75% and 100%) and 100 postlarvae stocked into each 800‐L tank. Salinity acclimation was done from 30 g/L to 6 or 1.5 g/L within 2–3 days by pumping water from adjacent shrimp production ponds. Following salinity acclimation, the S4 system maintained flow‐through at 1.5 g/L, while N10 system was maintained static at 6 g/L salinity. At the conclusion, no significant differences were observed for either survival or growth of shrimp postlarvae between RSS and LCSM treatments at all salinities examined. Results reflect the potential use of LCSM to replace RSS, which could be an excellent solution to bring down the cost of production in inland shrimp aquaculture.  相似文献
